Elspeth was born in 1555. Elspeth's father was Sir James Makgill and his mother was Jane Elizabeth Adamson. His paternal grandparents were James Makgill and Helen Wardlaw; his maternal grandfather was William Adamson, Burgess of Edinburgh and his maternal grandmother is Not Filled. He had five brothers and two sisters, named James Robert, John, Samuel, Zachary, David, Rachel and Elizabeth. He is the third youngest of the eight children.
