Samuel was born in 1547 in Rankeillour, Fife, Scotland. Samuel's father was Sir James Makgill and his mother was Jane Elizabeth Adamson. His paternal grandparents were James Makgill and Helen Wardlaw; his maternal grandfather was William Adamson, Burgess of Edinburgh and his maternal grandmother is Not Filled. He had five brothers and two sisters, named James Robert, John, Zachary, Elspeth, David, Rachel and Elizabeth. He was the third oldest of the eight children. He died at the age of 62 in 1609.
